Cancer Protein Description

This report provides a detailed description of a selected cancer protein with information collected from various sources, including UniProt, the Wellcome Trust Sanger Institute’s Catalogue of Somatic Mutations in Cancer (COSMIC), and the Atlas of Genetics and Cytogenetics in Oncology and Haematology.

Protein Name: DKK1
Gene Name: DKK1
Protein Full Name: Dickkopf-related protein 1
Alias: SK
Mass (Da): 28672
Number AA: 266
UniProt ID: O94907
Locus ID: 22943
COSMIC ID: DKK1
Gene location on chromosome: 10q11.2
Cancer protein type: TSP
Effect of cancer mutation on protein: LOSS
Effect of active protein on cancer: INHIBITS
Number of cancer specimens: 20237
Percent of cancer specimens with mutations: 0.34
Gene undergoes hypermethylation: DKK-1 is silenced in colon cancer by DNA hypermethylation and this correlates with advanced Dukes' stages of colorectal tumorigenesis
Normal role description: DKK1 is a secreted protein with two cys-rich regions that functions to inhibit Wnt signalling. It is involved in embryonic development and in vertebrate development, through local inhibition of Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. It is downregulated (by hypermethylation) in colon cancer.
