Cancer Protein Description

This report provides a detailed description of a selected cancer protein with information collected from various sources, including UniProt, the Wellcome Trust Sanger Institute’s Catalogue of Somatic Mutations in Cancer (COSMIC), and the Atlas of Genetics and Cytogenetics in Oncology and Haematology.

Protein Name: CFLAR
Gene Name: CFLAR
Protein Full Name: CASP8 and FADD-like apoptosis regulator
Alias: CASH; CASP8AP1; Caspase homologue; Caspase-eight-related; Caspase-like apoptosis regulatory; Caspase-related inducer of apoptosis; Casper; Cellular FLICE-like inhibitory; C-FLIP; C-FLIPL; C-FLIPR; CLARP; FADD-like anti-apoptotic molecule; FADD-like antiapoptotic molecule 1; FLAME; FLAME-1; FLIP; I-FLICE; Inhibitor of FLICE; MACH-related inducer of toxicity; MRIT; Usurpin; Usurpin beta
Mass (Da): 55344
Number AA: 480
UniProt ID: O15519
Locus ID: 8837
COSMIC ID: CFLAR
Gene location on chromosome: 2q33.1
Cancer protein type: OP/TSP
Effect of cancer mutation on protein: MIXED
Effect of active protein on cancer: MIXED
Number of cancer specimens: 19718
Percent of cancer specimens with mutations: 0.26
Transduced into viral genome: HHV-8
Normal role description: CFLAR has close homology to Caspase 8 but lacks caspase catalytic activity. Thus, CFLAR functions as a competitive inhibitor of the caspase pathway. CFLAR will therefore block Fas- and TNF-a mediate cell apoptosis. Howoever, some mutant variants have demonstrated oposite effect and trigger apoptosis.
